NoteInhibition of Proteasome Activity byTyropeptin A in
نویسندگان
چکیده
Tyropeptin A, a potent proteasome inhibitor not reported before, was produced by Kltasatospora sp. MK993-dF2. In this study, we inyestigated the effects of tyropeptin A on proteasome activity in PC12 cells. Tyropeptin A inhibited the intracellular proteasome activity in a dose-dependent way and seemed to cause neurite outgrowth. As expected, ubiquitinated proteins that should be substrates for the proteasome accumulated in cells treated with tyropeptin A. Hence, it appears that tyropeptin A can permeate into cells and there inhibit the intracellular proteasome actiyity.

The effect of a period of resistance training on the muscle proteasome activity and autophgy flux in mice with cancer induce cachexia
Introdution: Muscle weakness that is caused by cancer called Cachexia. One of the causes of the formation of the cachexia is the change in protein degradation, and the ubiquitous protease and Autophagy Lysosomes system is the most important protein breakdown system. Resistance training has been one the best stimulator of increasing muscular mass. Therefore, the purpose of this study was to inve...
متن کاملDesign, Synthesis and Evaluation of Substituted Aryl-2-Nitrovinyl Derivatives as Small Molecules Proteasome Inhibitors
Based on the existing structure activity relationship for proteasome inhibitors, a number of substituted aryl-2-nitrovinyl derivatives have been synthesized as Michael acceptor and their cytotoxicity and proteasome inhibitory effects were evaluated on two cancer cell lines. Compound 2d exhibited IC50 values of 0.71 and 17.79 μM comparable to bortezomib against MCF-7 and PC-3, respectively. The ...

Background: The ubiquitin-proteasome system (UPS) plays a crucial role in regulating the levels and functions of a large number of proteins in the cell, which are important for cancer cell growth and survival. The proteasome is highly activated in B-cell precursor acute lymphoblastic leukemia (BCP-ALL), which is the most common malignancy in children. The attempt to inhibit proteasome as a ther...

T-cell acute lymphoblastic leukemia is an aggressive hematologic malignancy which is usuallyassociated with unfavorable prognosis particularly in patients with refractory/relapsed disease. Therefore, development of novel therapeutic strategies is highly required for improving theoutcome of these patients. Although there are several studies evaluating the efficacy of proteasome<...
